(Macro)molecular engineering on the nanoscale

Samenvatting

The present proposal focuses on the design and preparation of (macro)molecules, assemblies, micelles, nanoparticles and nano-structured objects using well-defined building blocks in combination with covalent and non-covalent (supramolecular) bonds and interactions. For this purpose, living and controlled polymerization methods will be utilized in order to prepare suitable functionalized telechelics and (block) copolymers. The introduction of non-covalent binding motifs (e.g. terpyridine ligands) will either be performed on the level of the initiators and monomers or by end-group modification of the telechelics and (block) copolymers. Besides synthetic polymers, defined biopolymers, such as proteins or DNA, and nanoparticles will also be used. The state of association in solution will be of special interest in the course of the project in order to be able to understand and control in detail the self-organization and self-recognition processes. For this purpose, specific analytical methods from colloid and biophysical science will be used. In particular, analytical ultracentrifugation will be employed in order to study aggregation behavior and to determine the molecular weights. Furthermore, light scattering techniques and (cryo)TEM will also be considered. Moreover, preparative ultracentrifugation as well as preparative size exclusion methods will be applied to separate different assemblies and aggregates. Suitable nano- and micro-structured surfaces will be fabricated using a combination of chemical self-assembly processes and nano-lithography (based on scanning probe techniques). The selective and controlled build-up of functional nanostructures will be characterized by a combination of AFM, STM, NSOM and TEM with spectroscopic methods. In addition, scanning probe methods will also be used for the investigation of selected properties of the obtained systems (e.g. force spectroscopy, local conductivity measurements, etc.) and for manipulation on the nanoscale. Guided by the results of the characterization, the nanostructured systems and objects thus prepared will be evaluated with a view to potential applications.
